Scope
This standard specifies the method for determining the fatty acid content in food. This standard is applicable to the determination of total fat, saturated fat (acid), and unsaturated fat (acid) in food. The hydrolysis-extraction method in this standard is applicable to the determination of fatty acid content in food; the transesterification method is applicable to the determination of fatty acid content in oil samples with a free fatty acid content of no more than 2%.
Introduction

Core Essence of Standard Specifications

This testing standard is applicable to 12 types of food, including infant formula milk powder, animal and plant oils and fats, etc. It clearly uses the hydrolysis-ether extraction method as the basic pretreatment method, and integrates the original 7 testing methods for the first time.

Detection method decision tree

Method selectionApplicable scenariosFree fatty acid limit
Method 1 Internal standard methodAll food categoriesGeneral method
Method 2 Transesterification methodVegetable oil/animal fat≤2.0%
Method 3 Acetyl chloride methodMilk powder/anhydrous butterMoisture content <5%

Key nodes of process control

  • 100m poly(dicyandiamide) capillary column replaced the original glass column
  • Double buffered step temperature program (100℃-230℃/four-stage temperature control)
  • Precision control of Dow Corning pipette accuracy error ≤ 0.5μL

Standard Evolution Evaluation

Iteration highlightsMajor improvements
Chromatographic column upgradeSeparation efficiency increased by 34%/theoretical plate number reached 2000/m
Detection efficiency optimizationMaximum running time reduced to 82min
Quality control rigorQuantitative limit increased to 0.0033g/100g

High-risk reagent management specifications

15% boron trifluoride methanol must adopt an ice-salt water bath preparation system, and the acetyl chloride hydrolysis operation must be equipped with a chemical protective mask and splash guard. The pretreatment room should be equipped with a mechanical ventilation system with an air intake and exhaust rate of >30m³/min.

Matrix Application Guide

  • pH gradient hydrolysis process is preferred for dairy samples
  • For nut oils, refer to the FDA incremental month-on-month correction formula
  • EPA/DHA projects must be verified by photodiode detection
