Liquid chromatography (LC)

Steps:

  1. Sample preparation: Take 500 g of representative sample, add ascorbic acid and crush it.
  2. Extraction: Extract with n-hexane, concentrate with nitrogen and then dissolve.
  3. Chromatographic conditions: C18 column, acetonitrile-water (65:35), flow rate 1 mL/min, fluorescence detector.

Gas chromatography (GC)

Steps:

  1. Sample preparation: Same as LC method.
  2. Extraction: n-hexane extraction, nitrogen concentration and reconstitution.
  3. Chromatographic conditions: weak polar capillary column, nitrogen carrier gas, nitrogen phosphorus detector.

Implementation advice

Laboratory precautions:

  • Strictly control the purity and storage conditions of reagents, especially standards.
  • Instrument calibration: Regularly calibrate the fluorescence detector of the liquid chromatography and the nitrogen-phosphorus detector of the gas chromatography.
