cAMP是一种普遍存在于所有哺乳动物细胞中的第二信使。这种信使调节各种激素、神经递质、生长因子、细胞因子和其他配体受体的信号转导。cAMP磷酸二酯酶是水解和截断源自受体激活的cAMP信号传导的唯一途径。cAMP水解活性由一个相当大的多酶家族提供，该家族具有独特的细胞内定位和调节特性。在各种PDE家族（PDE1-PDE11）中，PDE4有4个基因（PDE4A、PDE4B、PDE4C、PDE4D）以多剪接变体形式表达。

PDE4活性测量需要两步反应，在第一步反应中，cAMP被PDE活性水解成非环状。通过使PDE源变性或加入PDE4选择性抑制剂来停止该反应，然后通过第二步酶反应将AMP转化为腺苷达到平衡。由于腺苷是中性的，因此通过离子交换色谱法用电荷分离cAMP或AMP。为了避免其他PDE的背景贡献，该PDE测定在针对PDE4优化的底物浓度下进行。PDE4酶活性与第一反应中形成的腺苷成正比。

PDE4活性测量在2-3小时内提供可重复的结果。该试剂盒提供足够的试剂，与内部控制一起处理50或100个样本。试剂盒试剂在推荐条件下储存可稳定6个月。该试剂盒的内容物必须按照瓶子标签和规格表上的指示在不同温度下储存。
